The Under-19s, who finished fourth in the league campaign after starting the season with six wins in a row, will face fifth-placed Atalanta at Sassuolo's Stadio Ricci - with kick-off on Tuesday set for 19:00 CEST.
There are no extra-time or penalties for this phase of the competition; should the match end in a draw after 90 minutes, Roma will progress by virtue of their higher finishing position in the league.
Sampdoria then await the winners of the tie in the semi-finals - with that game scheduled to take place on Saturday 26 June at 18:00 CEST.
Extra-time is in effect for the final two rounds of the competition, with the higher-seeded team only progressing if the semi-final tie also finishes level after 120 minutes.
A penalty shootout will be a possibility when it comes to the eventual final, however. That game will take place on Wednesday 30 June.
On the other side of the draw, Juventus and Empoli will face each other for the right to meet Inter Milan in the semi-finals.
