Zaniolo, 21, suffered a complete tear of the ACL in his left knee during Monday's UEFA Nations League game between Italy and Holland.
It is the second such setback for the forward - who had recovered from a similar injury to his right knee suffered back in January.
After an outpouring of support and messages on social media, Zaniolo took to Instagram to thank those who had shown concern.
"For the many, many people who have been asking how I am, this morning I underwent medical checks that revealed a tear of the cruciate ligament in my left knee," he wrote.
"I thank all of you, those of you who are Roma fan and those that aren’t, for the support… I will be back soon! Nicolo."
Zaniolo will undergo surgery on Wednesday at Villa Stuart private hospital in Rome.
