Spalletti, who spent two spells as head coach of the Giallorossi, could be in for a noisy reception at a sold-out Olimpico - but hopes the atmosphere will only serve to inspire his unbeaten side.
Napoli have won all eight of their Serie A games so far to sit top of the fledgling standings.
"Roma v. Napoli is my match - a match between two parts of me," Spalletti said at his pre-match press conference. "It's a hugely important game for Napoli's future."
He added: "Roma are an excellent team with an experienced coach. It certainly won't be an easy game. Mourinho raises the level of whichever league he is working in.
"This match could alter the course of the season for both teams. We must make sure we're 100% ready for it."
Almost 50,000 fans are expected to be at the Olimpico for the game, although Spalletti nonetheless believes his side will have to be focused on the 11 players his side will be up against rather than any hostile atmosphere.
"Tomorrow we need to make sure we are focused on the game, we need to be able to repel all Roma's attempts to attack," Spalletti said.
"They have some quality players, a dangerous attacking rhythm and power.
"They will try to attack the spaces wherever possible and we have to make sure we are stronger and more combative than they are."
