The Belgium international suffered a thigh injury while away on international duty last week, and returned to Rome to begin work on his recovery ahead of the game at the Stadio Olimpico.
And Di Francesco will wait to the last moment before making his decision.
"We will take a look at Nainggolan between today and tomorrow morning, and we will decide if he is available or not for the derby," Di Francesco said.
"Unlike a lot of players, he can be influential even when he is only at 80%. I will take that into account, but I don't want to pick him if he is less than 90%.
"I hope that between today and tomorrow he responds in the right way, because I would like to play him."
Di Francesco is no stranger to the passion of the Derby della Capitale, having experienced it during his playing days. And he will make sure his players go out on Saturday with the right attitude.
"We'll do everything we can to win the derby," Di Francsco added. "We have to give it our all, especially because the fans are showing us such great support.
"People are back at the stadium and we don't want to let them down."
